If your 2 stroke is running rich will that affect your compression

No, not directly.  Not unless you are so rich that liquid fuel is pooling in the cylinder causing a near hydrolock situation.  I have also seen some two strokes run so rich for such a long period of time, that the carbon build up causes the rings to get stuck and not seal well.  But that takes running it very rich for many hours.

Well, more oil in cylinder will give higher compression reading if rings are worn out.  Up to a point.  If your rings get clogged with carbon from running rich and stick, then compression will drop and cylinder could be damaged.
